Simultaneous LC–MS/MS measurement of urinary oxidative stress biomarkers

Simultaneous LC–MS/MS measurement of urinary oxidative stress biomarkers

Overview

At ADLM 2026, Eureka Lab Division, part of Sentinel Group, presented new data on the analytical performance of the OxiStress Urine LC–MS/MS kit, a quantitative panel designed for the simultaneous measurement of six urinary biomarkers associated with oxidative damage. The poster highlights the value of liquid chromatography–tandem mass spectrometry for the specific, standardized assessment of oxidative stress pathways in clinical and translational research.

Why oxidative stress biomarkers matter

Oxidative stress results from an imbalance between reactive oxygen and nitrogen species and antioxidant defenses, leading to cumulative molecular damage involving nucleic acids, lipids, and proteins. Urinary biomarkers reflecting these pathways—such as oxidized nucleosides, isoprostanes, and modified amino acids—are widely used because urine collection is non-invasive and can help capture systemic oxidative processes.

A unified LC–MS/MS urinary panel

The OxiStress Urine LC–MS/MS kit enables the simultaneous quantification of six complementary biomarkers within a single analytical workflow: DNA/RNA oxidation markers (2-dG, 8-OHdG, 8-OHGuo), lipid peroxidation marker 8-PGF2α, and protein oxidation/nitration markers DiY and 3-NT. By consolidating these markers into one method, the panel supports a broader assessment of oxidative status compared with conventional single-analyte approaches.

Analytical performance

The method showed excellent chromatographic separation, stable retention times, and high linearity across the evaluated calibration range, with correlation coefficients consistently above 0.995. Precision data confirmed robust analytical performance, with within-run and between-run coefficients of variation below 17% and low limits of quantification for all six biomarkers. These results demonstrate the potential of the OxiStress Urine LC–MS/MS kit as a sensitive and reliable approach for simultaneous quantification of urinary oxidative-stress biomarkers.

The value

The LC–MS/MS approach may facilitate comprehensive multi-marker assessment of oxidative stress in sports medicine, wellness, and broader clinical or translational research applications. The availability of a unified urinary panel can help laboratories investigate complementary oxidative pathways through a single-run workflow with high analytical specificity.
