Validation of a New Freeze-Dried PCR Universal Master Mix
AACC 2010, Anaheim - California

M. Gramegna, E. Castiglioni, A. Moiana, M. Zanussi, M. Ferrari
Sentinel Ch. has recently developed a new proprietary freeze-drying system for the manufacturing of ready-to-use PCR Universal Master Mixes. Usually the PCR mixes need to be stored at -20°C or + 2/8 °C. This means request for refrigerators and freezers in the labs, and controlled temperature in case of shipment. The new system is able to maintain the activity of the mix with no loss of performance at room temperature. The aim of the present study is to validate the performances of the freeze-dried mixes for the main molecular applications in research or diagnostic laboratories.

CRP Vario – An Immunoassay for Ultrasensitive and Wide-Range Assay of CRP on AEROSET® and ARCHITECT®

C-Reactive Protein (CRP) is the best-known among the acute-phase proteins. CRP concentration increase in response to inflammation, postoperative conditions or tissue damage. CRP normally present in low concentration (< 5 mg/L) can raise rapidly during the acute inflammatory process with values up to 500 mg/L during bacterial infections. In recent years, studies have shown that in apparently healthy individuals a direct correlation between the level of CRP and the risk of developing coronary heart disease (CHD) exists.
CRP has also been reported as a potential marker for monitoring Statin therapy. The various clinical applications of CRP require access to both a sensitive and a wide range assay.
CRP Vario was designed to fulfill both requirements.

STUDY OF SAMPLE-TO-SAMPLE AND REAGENT-TO-REAGENT ON AEREOSET® AND ARCHITECT®

Modern random access analysers are playing a determining role in the productivity of clinical laboratories and their ability to offer clinicians an ever-wider range of tests with state-ofthe-art analytical performance. Assay sensitivity increasing and the number of on-board reagents expanding, however, the potential for interaction among and between samples, reagents and analyzer has increased, hence, the possibility for a negative impact on patient results. We here report on our brief to verify sample-tosample and reagent-to-reagent carry over for a total of 32 new reagents equating 32 x (31+ 54) x 2 or 5440 reagent combinations on the Abbott AEROSET® with confirmation of carry over on ARCHITECT® c8000 clinical chemisty analyzers.

COMMUTABILITY OF 17 SENTINEL REAGENTS FOR USE ON AEROSET® AND ARCHITECT®

Depending on the individual clinicallaboratory needs it is common practice for instrument manufacturers to offer the choice of more than one analyser. These instruments can be entirely different or more often belong to the same family, differing only in terms of capacity and specific performance characteristics.
The goal of this study was to evaluate equivalency of the AEROSET® and ARCHITECT® c8000 (Abbott). Using a validation protocol created for this specific purpose we have compared the performance of 17 IVD products (Sentinel CH, Milan, Italy) on the 2 instruments with regard to precision, accuracy and cross-interference.

EXPANSION OF ABBOTT ANALYZERS REAGENT MENU: VALIDATION OF 17 SENTINEL REAGENTS

New tests previously performed using alternative methods (e.g. Atomic Absorption, Nephelometry, Potentiometry) on different instruments, the necessity for clinical laboratories to overhaul and improve their services in terms of turnaround time, breadth and depth of test menu, quality of results, and in general new value added, together with dwindling economic resources and the availability of high-throughput random-access analysers integrating in-line immunochemistry testing modules have significantly changed the nature of laboratory work. Work is no longer strictly partitioned between diagnostic areas and an increasing number of lab activities are being merged. To address this challenge Sentinel and Abbott’s scientific and technical work teams have joined forces. We here report on the outcome of this collaboration for the optimisation of 17 in-vitro diagnostic products (Sentinel) and their validation for routine use of the Abbott AEROSET® and ARCHITECT® c8000 analysers according to the requirements of the European Directive 98/79/CE.

C-Reactive Protein (CRP) is the bestknown among the acute-phase proteins, the concentration of which increases in response to inflammation, postoperative conditions or tissue damage. CRP is normally present in low concentration (<5 mg/L) but increases rapidly during the acute inflammatory process with values increasing up to 500 mg/L during bacterial infections. The level of CRP can be detected 6 hours after inflammation starts reaching a peak at 48 hours. The determination of CRP represents a useful laboratory test for the detection of acute infection and for monitoring of inflammation in acute rheumatic and gastrointestinal diseases. In recent years, studies have shown that in apparently healthy individuals a direct correlation between the level of CRP and the risk of developing coronary heart disease (CHD) exists.
